On July 24, 2011, Mendy signed his first three-year contract with Ligue 2 club Le Havre. He then made his professional debut in the team's 2–1 Coupe de la Ligue second-round defeat to Amiens on August 9, 2011.

On 8 July 2013, Mendy signed with Ligue 1 side Olympique de Marseille. The same year on August 11, he made his club and Ligue 1 debut against Guingamp. In his second season with the Marseille, he was included on the 40-man shortlist for the "2014 Golden Boy Award".

On June 22, 2016, Mendy signed a five-year contract with Olympique de Marseille's Ligue 1 rivals Monaco. He made his debut for the club in the first leg of the Champions League third-round match against Fenerbahçe on July 27.

 

On March 1, 2017, Mendy scored his first goal for Monaco in the Coupe de France against former club Marseille. He was a key part of Monaco's 2016–17 Ligue 1 championship season as he started for the team in 24 matches under Portuguese football manager Leonardo Jardim.

On July 24, 2017, Mendy moved to Premier League side Manchester City signing a 5-year contract for a reported fee of £52 million.

 

On September 29, Mendy playing against Crystal Palace, sustained an injury, hurting his right knee following a challenge against Palace forward English professional footballer Andros Townsend.

 

Mendy underwent surgery in Barcelona on September 29 and returned from injury on April 22, 2018, as a 75th-minute substitute for English professional footballer Fabian Delph in a game against Swansea City at the City of Manchester Stadium. Mendy has played 4 games for the club to date.
