Mary-Louise Parker was born on August 2, 1964, in Fort Jackson, South Carolina, to parents Caroline Louise (née Morell) and John Morgan Parker. Her father was a judge and her mother was a nurse. She has two siblings, a brother, and a sister.

Parker attended the North Carolina School of the Arts, where she studied acting. She graduated in 1986 with a Bachelor of Fine Arts degree in drama. After graduation, she moved to New York City to pursue an acting career.
