Sidse Babett Knudsen was born in Copenhagen, Denmark, on 22 November 1968 to Ebbe Knudsen, a photographer, and Susanne Andersen, a school teacher. She holds Danish nationality and regarding her ethnicity, she is Germanic. She grew up in her hometown Copenhagen along with two brothers named La s se Knudsen and Niclas Knudsen.
Knudsen took acting lessons from 1987 to 1990 at the Theatre de l'Ombre in Paris. Upon coming back to Denmark, she worked for the experimental theatre OVINE 302. She also performed at the Betty Nansen and Royal Danish Theatres in Copenhagen.

Sidse Babett Knudsen landed her first role in 1997 appearing in Jonas Elmer's slice-of-life comedy Let's Get Lost. For her breakout performance in the movie, she went on to win several accolades such as Robert Award and the Bodil Award for Best Actress.
